Will Programs work after rename the drive letter

We have one physical server running Windows 2003. There are two drives (C and D) on one disk. C is out of the spaces. We plan to add iSICS drive as E drive to the server. Use robocopy copy all folders and files from D to E. Delete the D and rename the E to D. Extend C drive using the unallocated space on the internal disk. There are some programs and SQL data on the D drive. Will the programs and SQL data work after copying and re-assigning D letter?

Based on my test, if you move the program file to another disk via robocopy, you still can run the program in our server. Here is my test:

1. On the Windows Server 2003, add two disk(C: and D:) and iSICS disk.

2. Install an program Bing Input on the D: drive, then use robocopy to iSSISC disk.

3. Then delete the D: and rename the iSISC disk as D: driver, then I find that the program still can use. But the shortcut cannot be used. We should re-create a new shortcut of the program.
