Q: Should we disable IPv6 DNS if we don't use it and our Router doesn't setup IPv6?

A: Disabling or changing default IPv6 behavior is not recommended mainly because it is not the mainstream tested code path. Therefore, having IPv6 enabled will result in the safest configuration for your production deployment. And other products also rely on IPv6, such as Remote Assistance, HomeGroup, DirectAccess, Windows Mail.

That being said, you could change the preference to have IPv4 preferred over IPv6 without disabling IPv6 completely:
